Which Products do you need?

How do the products fit into the lentivirus packaging workflow?

The Packaging System Mix (1) and the DNAfectinTM Plus Transfection Reagent (2) are required to generate the lentivirus during transfection of the cells with the transfer plasmid. After collection, the virus concentration can be measured (titered) using the qPCR Lentivirus Titer Kit (3). To improve transduction (infection) efficiency, the ViralEntryTM Transduction Enhancer (4) can be added along with the virus particles directly to the cells and media.

2nd Generation vs. 3rd Generation Packaging System Mix

The 2nd Generation Packaging System Mix is recommended unless your facility requires 3rd Generation Packaging Systems for stricter safety regulations.

Feature 2nd Generation Packaging System Mix 3rd Generation Packaging System Mix
Transfer Plasmid Can package 2nd or 3rd Generation transfer plasmids (Tat dependent or independent) Can ONLY package 3rd Generation transfer plasmids (Tat independent)
Packaging Plasmids One plasmid encoding Gag, Pol, Rev, and Tat Two plasmids: one encoding Gag and Pol, and one encoding Rev
Envelope Plasmid Interchangeable: usually encodes for VSV-G Interchangeable: usually encodes for VSV-G
Safety Safe. Replication incompetent: Uses 3 separate plasmids encoding various HIV genes. Safer. Replication incompetent and always self inactivating: Uses 4 plasmids instead of 3 and eliminates the requirement for Tat.
Recombinant Virus Yield Higher yields than 3rd Generation due to fewer plasmids (3) required to be transfected Lower yields than 2nd Generation due to more plasmids (4) required to be transfected
